Genetic transformation including vector construction

Drosophila embryo transformation to generate transgenic lines requires special equipment and technical expertise. In many cases it is more time and cost effective to request this service from external sources.

Aktogen Limited offers a range of services for professionals that includes:

a. custom made genetic vector construction
b. embryo injection
c. isolation of transgenic fly strains

Please note that we are considering to add genetic transformation using the PhiC31 integrase system to our services.

 We assume for the P-element transformation services that
1.    the customer sends a clean DNA sample (15 ml of 1mg/ml DNA – purified e.g. with Endotoxin-free Qiagen gravity column –, 0.25mg/ml helper plasmid)
2.    the P-element construct is not larger than 13 kb (normal transformation rate)
3.    the genetic construct is not lethal
4.    in these – majority – of situations we guarantee 3 independent transformants per construct 

If the DNA sent by the customer is lethal (no survival after injection) we inject 200 embryos once again for free to minimize the chance that lethality is due to injection error.

If the genetic construct sent by the customer is lethal (no transformant but many survival after injection) we inject 200 embryos once again for free to minimize the chance that the low transformation rate is due to injection error.
